Design Notes
Designed for minimal basal transcription by altering binding site sequence and distance. The RBS was specifically designed using Vogit lab RBS calculataor[http://voigtlab.ucsf.edu/software/] to work in the context of enhanced hyperspank promoter and BBa_K316012. The promoter and RBS are coding on the negative strand to minimise read-through from upstream sequences.
